ABSTRACT

In recent years, with the change in life style, social environment, and national childbearing policy, the proportion of high-risk pregnant women has increased significantly, triggering the spectrum of obstetric diseases to constantly change, which has brought new challenges to the diagnosis and treatment of obstetrics. Traditional Chinese medicine (TCM) has been proved effective in dealing with a variety of obstetric diseases, and various treatment methods are available, which can serve as alternative means for solving refractory obstetric diseases. However, most obstetric clinicians are currently less aware of the therapeutic effects of TCM, which has significantly hindered its participation in clinical treatment. Therefore, the China Association of Chinese Medicine (CACM) organized the outstanding young obstetricians of TCM and western medicine to discuss 15 obstetric diseases responding specifically to TCM or integrated TCM and western medicine, including hyperemesis gravidarum, threatened abortion, ectopic gestation, cough during pregnancy, pregnancy-induced hypertension syndrome, maternal-fetal ABO incompatibility, postpartum hypogalactia, residual pregnancy tissue in uterine cavity, puerperal infection, pantalgia after childbirth, hematoma/undesirable healing after caesarean section, postpartum urinary retention, ileus after cesarean section, pelvic floor dysfunction, and postnatal depression. The suggestions for their treatment with TCM or integrated TCM and western medicine were also proposed, aiming to provide patients with effective and personalized treatments in clinical practice and improve the diagnosis and treatment effects of obstetric diseases, thus benefiting the public. At the same time, more obstetrical clinicians are expected to understand the therapeutic effects and advantages of TCM and draw on the strengths of both TCM and western, thereby promoting the establishment of an obstetric diagnosis and treatment system with Chinese characteristics.

ABSTRACT

Objective To explore the effects of problem-based learning (PBL) method on the efficiency of teaching of Chinese medicine dermatology based on medical records. Methods Undergraduate students who were in the same grade were divided into experimental group and control group. The experimental group was taught with the PBL method based on medical records. Teachers selected medical records, combed knowledge points, designed questions and carried out teaching in response to questions. The control group was taught with traditional teaching method. The teaching efficiency of two groups was evaluated by using test scores and questionnaire survey. Results The scores of syndrome differentiation scores, medication scores, and the overall score of experimental group were better than those in the control group. Compared with the control group, learning initiative, ability to involve in the teaching and master knowledge, and ability of logical analysis and communication of experimental group were all improved remarkably. Conclusion PBL method based on medical cases is beneficial for the improvement of the level of diagnosing and treating skin diseases based on syndrome differentiation and ability of solving problem.

ABSTRACT

Objective To clone the R2R3 MYB transcription factor gene SmMYB87 in subgroup 14 from Salvia miltiorrhiza, and analyze the bioinformatics and expression of this gene. Methods Total RNA extracted from S. miltiorrhiza was used as cDNA synthesis template and the full length cDNA sequence was obtained through homology-based cloning and rapid amplification of cDNA ends (RACE) technology. The structure and physicochemical properties of SmMYB87 gene and its coded protein were analyzed with bioinformatics softwares. The expression of SmMYB87 in different organs was determined with qRT-PCR, and a GFP fusion expression vector was constructed to investigate the subcellular laicization of SmMYB87 protein. Results SmMYB87 gene contained two introns and an open reading frame (ORF) of 732 bp, encoding 243 amino acid polypeptides. It expressed in roots, stems, leaves and flowers with similar expression levels and the SmMYB87 protein located in nucleus and cytomembrane. Conclusion The analysis of sequence structure and expression pattern of SmMYB87 will be helpful to study the regulating roles of this gene in S. miltiorrhiza.

ABSTRACT

This study was designed to investigate the effect of Xiao-Ai-Ping injection on cancer angiogenesis. CCK8 assay and BrdU incorporation immunofluorescence assay were used to detect the effect of Xiao-Ai-Ping injection on HUVECs proliferation; wound healing assay and transwell assay were employed to test the effect of Xiao-Ai-Ping injection on HUVECs migration. The anti-angiogenic effect of Xiao-Ai-Ping injection was examined by tube formation assay, rat aortic ring assay and chicken chorioallantoic membrane (CAM) assay. ELISA assay was used to measure the secretion of vascular endothelial growth factor (VEGF); and the activation of vascular endothelial growth factor receptor 2 (VEGFR2) protein and its downstream signaling pathways were examined by Western blot. Our data demonstrated that Xiao-Ai-Ping injection inhibited HU-VECs proliferation in a time-and dose-dependent manner, and the IC50 (mg·mL-1) values for 24, 48 and 72 h were 48.7±7.14, 29.1±2.25 and 22.0±4.53, individually. Xiao-Ai-Ping injection inhibited HUVECs DNA synthesis and migration. Xiao-Ai-Ping injection suppressed HUVECs tube formation, and reduced microvessel sprouting from rat aortic rings and vessel growth in CAMs. Furthermore, Xiao-Ai-Ping injection attenuated the secretion of VEGF, and inhibited the expression of p-VEGFR2 and phosphorylation of protein kinase B (p-AKT). We conclude that Xiao-Ai-Ping injection inhibits angiogenesis by down-regulation of VEGF signaling and AKT pathway.

ABSTRACT

Marsdenia tenacissima extract (MTE, trade name: Xiao-Ai-Ping injection) is an extract of a single Chinese plant medicine. It has been used for the treatment of cancer in China for decades, especially for esophageal cancer and other cancers in the digestive tract. In the present study, the potential mechanism for MTE's activity in esophageal cancer was explored. The effects of MTE on the proliferation of human esophageal cancer cells (KYSE150 and Eca-109) were investigated by the MTT assay, the BrdU (bromodeoxyuridine) incorporation immunofluorescence assay, and flow cytometric analysis. MTE inhibited cell proliferation through inducing G0/G1 cell cycle arrest in KYSE150 and Eca-109. Western blot analysis was employed to determine protein levels in the MTE treated cells. Compared with the control cells, the expression levels of the cell cycle regulatory proteins cyclin D1/D2/D3, cyclin E1, CDK2/4/6 (CDK: cyclin dependent kinase), and p-Rb were decreased significantly in the cells treated with MTE at 40 mg·mL(-1). In addition, MTE had an inhibitory effect on the MAPK (mitogen-activated protein kinase) signal transduction pathway, including ERK (extracellular signal-regulated kinase), JNK (c-Jun N-terminal kinase), and p38MAPK. Moreover, MTE showed little additional effects on the regulation of cyclin D1/D3, CDK4/6, and p-Rb when the ERK pathway was already inhibited by the specific ERK inhibitor U0126. In conclusion, these data suggest that MTE inhibits human esophageal cancer cell proliferation through regulation of cell cycle regulatory proteins and the MAPK signaling pathways, which is probably mediated by the inhibition of ERK activation.

ABSTRACT

Our previous research indicated that the Streptomyces pactum Act12 (Act12) had a certain promotional effect on tanshinone accumulation and up-regulated the expression of genes 3-hydroxy-3-methyglutaryl-CoA reductase (HMGR) and 1-deoxy-d-xylulose-5-phosphate reductoisomerase (DXR) in Salvia miltiorrhiza hairy roots. This study focuses on the roles of reactive oxygen species in S. pactum Act12-induced tanshinone production in S. miltiorrhiza hairy roots. The 4% Act12, 4% Act12 + CAT and 4% Act12 + SOD were added to S. miltiorrhiza hairy root and subcultured for 21 days, the dry weight, contents of reactive oxygen species, contents of tanshinones and expression of HMGR and DXR were determined at different harvest-time. The generation of reactive oxygen species (ROS) in S. miltiorrhiza hairy roots was triggered by 4% Act12 treatment. The relative expressions of genes HMGR and DXR in 4% Act12 treatment were 32.4 and 4.8-fold higher than those in the control. And the total tanshinone in the hairy roots was 10.2 times higher than that of the control. The CAT and SOD could significantly inhibit the ROS accumulation and relative expressions of genes HMGR and DXR in 4% Act12 treatment, which induced the total tanshinone content was decreased by 74.6% comparing with the 4% Act12 treatment. ROS mediated Act12-induced tanshinone production. The Act12 may be via the ROS signal channel to activate the tanshinone biosynthesis pathways. Thereby the tanshinon content in hairy roots was increased.

ABSTRACT

Objective To evaluate the clinical efficacy and safety of Chushi pellets in patients with subacute eczema of more dampness less heat type.Methods A randomized,double blind,double-dummy and positive parallel-controlled study was performed.Totally,129 patients with subacute eczema were collected at the dermatology clinic in Beijing Hospital of Traditional Chinese Medicine from May 2010 to October 2011,and classified into two groups to be treated with Chushi pellets and placebo capsules (n =66,experiment group) or Shiduqing capsules and placebo watered pills (n =63,control group) for 4 weeks.Follow-up visits were scheduled on day 1 (baseline),and 4 weeks after the beginning of treatment.Clinical endpoints included skin rash area,color,morphology,degree of pruritus and laboratory examination results.The therapeutic effect and safety of Chushi pellets were evaluated.The differences in clinical parameters were assessed by using t test and Wilcoxon rank-sum test between pre-and post-treatment,and by Pearson chi-square test or Fisher's exact test between the experiment and control group.Results Finally,the treatment outcome of 123 patients were assessed for the efficacy of Chushi pellets.At the end of the treatment,the total response rate was 69.3% (43/62) in the experiment group,and 59.0% (36/61) in the control group (x2 =1.43,P > 0.05).The improvement in pruritus was greater in the experiment group than in the control group (x2 =4.05,P < 0.05).Side effects mainly included loose stool and stomach upset,with an incidence of 13.6% (9/66) in the experiment group,and 11.1% (7/63) in the control group.No marked changes were observed in laboratry parameters after treatment in either of the groups.Conclusions Chushi pellets are safe and effective for the treatment of subacute eczema of more dampness less heat type.Compared with Shiduqing capsules,Chushi pellets shows an equivalent overall efficacy but a stronger antipruritic effect.

ABSTRACT

<p><b>OBJECTIVE</b>To establish a HPLC-ELSD fingerprint of Marsdenia tenacissima from different habitats, in order to provide a reliable method for scientific assessment and effective quality control.</p><p><b>METHOD</b>HPLC-ELSD was adopted to determine 25 baches of M. tenacissima herbs from different habitats. Traditional Chinese medicine chromatographic fingerprint similarity software assessment system 2004 developed by China Pharmacopoeia Committee was adopted to establish a common mode chart and assess chromatographic similarity based on the degree of correlation.</p><p><b>RESULT</b>The common mode for M. tenacissima herb C21 steroidal fingerprint was established, including 11 common characteristic peaks. Among them, 10 were identified. According to the assessment on the similarity of 25 batches of samples, 80% of them showed a similarity of over 0.80 in steroidal HPLC-ELSD fingerprint.</p><p><b>CONCLUSION</b>The method can be used to assess the quality of M. tenacissima.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To investigate and compare the efficacy and safety of two kinds of Qinbai ointment prepared conventionally and finely (C-QBO and F-QBO) in treating psoriasis in the active stage of blood-heat syndrome type, and to observe their influences on vaginal epithelial cell mitosis, also on keratinization of caudal flakes in mice.</p><p><b></b>METHODS</p><p><b>CLINICAL OBSERVATION</b>Adopting randomized, single-blinded, controlled design, 93 patients administered orally with Liangxue Huoxue Decoction were randomized into three groups treated externally with C-QBO, F-QBO and white vaseline respectively, applied twice a day for 8 weeks. The safety was checked and the changes of modified psoriasis area severity index (PASI), as well as the conditions of skin lesions (size, erythema, infiltration, squama, and itching) and symptom improving time were compared. Experimental study 1: Mice were randomized into the model group, the C-QBO group, the F-QBO group and the blank group, 7 in each group. Their vaginal epithelial cell mitosis indices were compared after 3 days of treatment with corresponding remedies. Experimental study 2: Mice were randomized into the C-QBO group, the F-QBO group and the blank group, 7 in each group. After the mice had been treated with corresponding remedies for 28 days, the granular layer formation in their caudal scales was compared.</p><p><b></b>RESULTS</p><p><b>CLINICAL OBSERVATION</b>The markedly effective rate was 63.3% (25/30) in the C-QBO group, 66.7% (20/30) in the F-QBO group, and 36.7% (11/30) in the control group. No statistical difference was showen between the two QBO treated groups (P > 0.05), but that in both of them was significantly different from that in the control group (P < 0.05). PASI scores lowered after treatment in all the three groups (P < 0.01), but the improvement in the two QBO groups was better than that in the control group (P < 0.05); the conditions of skin lesion were improved in all groups, but the improvements were more significant in the two QBO groups in terms of squama, infiltration and itching (P < 0.05), in aspects of improving time on erythema, infiltration and itching, especially the itching, F-QBO was superior to the C-QBO. Experimental study 1: The mitosis index in both QBO groups was lower than that in the blank group and the model group (P < 0.01). Experimental study 2: Number of scales with granular layer formation was higher in the two QBO groups than in the blank group (P < 0.01).</p><p><b>CONCLUSION</b>C-QBO and F-QBO can effectively relieve the skin lesion of psoriasis patients in the active stage of blood-heat type, and they could also promote the formation of epithelial granular layer in the caudal scales of mice.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To explore the feasibility and safety of conditioning regimen containing fludarabine (Flud) for haploidentical hematopoietic stem cell transplantation (HSCT).</p><p><b>METHODS</b>Preparative regimen containing Flud 40 mgxm(-2)xd(-1) on day -7 to -3 in place of cyclophosphamide (CTX) for haploidentical HSCT was given to 35 patients with hematologic malignancies (4 standard risk, 16 high risk, 15 relapse with no remission). All donors received rhG-CSF followed by HSC harvest. One patient received peripheral blood HSCT (PBSCT), one bone marrow transplantation (BMT), and the others BM combination with PBSCT. The regimen-associated side effect, engraftment, incidence of graft-versus-host disease (GVHD) and disease-free survival (DFS) probabilities were observed.</p><p><b>RESULTS</b>All patients achieved sustained, full donor-type engraftment. Thirty-four patients obtained primary durable engraftment, and 1 who rejected graft from his mother obtained successful durable engraftment after the second graft from his father. The cumulative incidence of grade III-IV acute GVHD and chronic GVHD was 12.1% and 31.7%, respectively. With a follow-up duration of 8-25 months, 6 patients were dead, in which 3 died of relapse, 2 of acute GVHD, 1 of fungal infection, none died of regimen-associated side effect. The other 29 patients remained alive and DFS probability was 79.7%.</p><p><b>CONCLUSION</b>Flud based conditioning regimens for haploidentical HSCT is safe and feasible, which reduces regimen-associated side effect, with no increasing the rate of relapse and infection, and decreases the incidence of aGVHD.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To survey the married couples at reproductive age for contraceptive knowledge in 5 Chinese cities.</p><p><b>METHODS</b>The data derived from a questionnaire-based research conducted in 2002 on family planning among married couples in 5 Chinese cities were statistically analyzed.</p><p><b>RESULTS</b>The total rate of good contraceptive knowledge was 70.10% in the couples. Among all the contraceptive measures, condom use was the most familiar one. The understanding of contraceptive knowledge in couples working in official departments and educational institutes was better than that among the couples working in common enterprises, affected by such factors as gender, geographic regions, educational background and willingness of child-bearing.</p><p><b>CONCLUSION</b>The married couples in the Chinese cities have generally good understanding of contraceptive knowledge, which can be affected by different factors among different populations.</p>

ABSTRACT

Orjective:To obtain recombinant CHO-K1 with expressing sPDGFR? and to identify the biological activities of sPDGFR? secreted in non-serum medium.Methods:Recombinant human sPDGFR? expression vector pIRES-Neo3-sPDGFR?-Fc was constructed and then transfected into CHO-K1 cells by using LipofectamineTM 2000.After screened with G418 in 8 weeks,some monoclone cells were selected randomly to amplify in 96-well-plate to 24-well-plates,and then to identify positive cell clones by RT-PCR.Furthermore,the candidate cell clones were test by Real-Time PCR and Western blot assays.Finally,anti-proliferation activities of the expressed sPDGFR? were analyzed by MTT.Results:sPDGFR?-Fc was cloned into pIRES-Neo3 correctly.The sPDGFR?-Fc expression level in recombinant CHO-K1 cell clones were concordant in between Realtime PCR and Western blot assay.sPDGFR?-Fc obtained from cultured non-serum medium of positive CHO-K1 could significantly inhibit proliferation of vascular endothelial cell.Conclusion:Successed to select recombinant CHO-K1 cell lines with high expressed sPDGFR?-Fc.The sPDGFR?-Fc can inhibit the cell proliferation significantly and it means sPDGFR?-Fc might be a new anti-cancer drug in the future.

Based on syndrome differentiation,other methods are used,such as clearing method,warming method,tonifying method,and drugs of promoting blood flow,promoting granulation and astringing are used locally.

ABSTRACT

<p><b>OBJECTIVE</b>Allogeneic marrow transplantation is a curative therapy for thalassemia, but no more than 30% of patients have HLA-indentical sibling marrow donor. The selection of alternative donors of unrelative marrow and the study on the probability of treating thalassemia major with unrelated donor bone marrow transplantation are of importance.</p><p><b>METHODS</b>Nine children with thalassemia were included in the study, and their gene mutational type were homozygote of thalassemia and double heterozygote, respectively. All of them were finally diagnosed of thalassemia major, and treated with unrelated donor bone marrow transplantation. To high-resolution HLA typing, two patients were matched, five had one unmatched isoform and two had two unmatched isoforms. The erythrocyte blood type was not matched in six patients. The preparative regimen included busulfan (oral use, 16 mg/kg, divided for 4 days), cyclophosphamide (intravenous use, 200 mg/kg, divided for 4 days), antithymocyte immunoglobulin (intravenous use, 30 mg/kg, divided for 3 days), and fludarabine (intravenous use, 125 mg/m(2), divided for 3 days). Ciclosporin A and methotrexate were used for graft-versus-host disease (GVHD) prophylaxis.</p><p><b>RESULTS</b>All patients had allergen reactions. One had hypotension. Five patients experienced I degrees approximately III degrees acute GVHD in the skin, while one had II degrees acute GVHD in liver. One patient had III degrees GVHD of intestines and gradually developed chronic GVHD in the skin, lungs and brain. One patient died of pulmonary hemorrhage. The duration when peripheral blood neutrophil count exceeded 0.5 x 10(9)/L was 12 - 26 days. The recovery time of WBC was as long as 23 - 110 days. Thrombocytes exceeded 50 x 10(9) within 61 approximately 142 days. The time when hemoglobin reached 100 g/L varied from 23 to 116 days. The last blood transfusion was on 13 - 62 days. Eight patients were fully grafted, while one was not grafted. During the 6 - 24 months of follow-up, seven patients' genotype of thalassemia major became normal. The erythrocyte blood type of five patients also changed into the same as that of donor. The hemoglobin was kept over 110 g/L without blood transfusion.</p><p><b>CONCLUSION</b>The transplantation of unrelated donor bone marrow for thalassemia major was successful. Unrelated donor bone marrow transplantation could cure thalassemia major, which expanded the marrow donor source for the transplantation of thalassemia major.</p>
